Actin monomer sequestering

The identified molecular function of thymosin β4 is sequestration of G-actin. The dissociation constant of the complex sits in the micromolar range, which allows fast binding and release, and β-thymosins are described as the main intracellular G-actin-sequestering peptides in most cells. This is the one activity with an explanation grounded in a defined molecular interaction.

Cytoskeletal and motility readouts

Overexpression or addition of exogenous thymosin β4, or its homologue β10, alters the actin cytoskeleton and has multiple effects on functions related to cell motility. Reviews note that some of these effects are consistent with a monomer-binding protein acting alone and others are not, leaving direct and indirect contributions unresolved.

Vascular and fibrotic models

In preclinical cardiac ischaemia models, treatment reduced infarct volume and preserved function, with antifibrotic and proangiogenic activity reported separately from infarct size.
