KPV (Lys-Pro-Val) is a tripeptide frequently referenced as an α-MSH fragment (11–13) in experimental literature. Due to its size and stability relative to longer peptides, KPV is commonly used as a research peptide when evaluating peptide signaling hypotheses tied to melanocortin-adjacent pathways and immune/inflammation signaling markers in controlled assays. Nuclear factor κB

In immortalised human bronchial epithelial cells the tripeptide produced dose-dependent inhibition of NF-κB signalling evoked by TNFα and respiratory syncytial virus, alongside reduced matrix metalloproteinase-9 activity and lower IL-8 and eotaxin secretion. The effect was associated with nuclear import of the peptide, stabilisation of IκBα and suppressed nuclear translocation of p65RelA, with competition assays indicating an interaction at the importin-α3 binding site.

Peptide transporter uptake

Work in intestinal models describes uptake of the tripeptide through the PepT1 di- and tripeptide transporter, with reduced inflammatory readouts reported downstream of that route. This is the basis on which the fragment is examined separately from the parent hormone: it enters cells by a transporter that longer melanocortin peptides do not use.

α-MSH fragment context

The sequence corresponds to residues 11-13 of α-melanocyte-stimulating hormone. Reviews of the parent hormone place the fragment among the C-terminal sequences that retain activity in inflammatory readouts, and report inhibitory activity for both α-MSH and KPV against Staphylococcus aureus and Candida albicans in antimicrobial assays.

The record for this tripeptide is preclinical and largely in vitro. Much of it sits within the wider literature on α-MSH rather than on the fragment alone, and the compound holds no FDA approval.
