SLU-PP-332 is a synthetic small molecule that activates all three estrogen-related receptors (ERRα, ERRβ and ERRγ), most potently ERRα. These orphan nuclear receptors regulate genes of mitochondrial biogenesis, oxidative phosphorylation and fatty acid oxidation, and the compound is studied as an exercise mimetic in mice.

Designing ERRα agonists had proved difficult; SLU-PP-332 came from work converting an ERRβ/γ-selective scaffold into pan agonists. It increased mitochondrial function and respiration in a skeletal muscle cell line, and in mice it increased type IIa oxidative fibres and exercise endurance through an ERRα-dependent acute aerobic exercise programme.

In diet-induced obese and ob/ob mice it increased energy expenditure and fatty acid oxidation and reduced fat mass accumulation, with improved insulin sensitivity. Pan-ERR agonists improved cardiac fatty acid metabolism and mitochondrial function in a pressure-overload heart failure model, and in aged mice the compound reversed mitochondrial dysfunction and inflammation in the kidney.

The group that developed it reports that SLU-PP-332 lacks oral bioavailability; in their comparison it was given intraperitoneally, and a chemically distinct successor, SLU-PP-915, was made to be orally active. Anti-doping laboratories have characterised its in vitro metabolites, since exercise mimetics are prohibited in sport.
