Epithalon (also written Epitalon; Ala-Glu-Asp-Gly, AEDG) is a synthetic tetrapeptide designed from the amino-acid composition of Epithalamin, a peptide extract of bovine pineal gland. It is studied mainly in ageing research, for reported effects on telomerase, melatonin synthesis and gene expression.

The tetrapeptide was designed from the pineal preparation Epithalamin, which in the originating group's work increased melatonin production in rats and lifespan in rats, mice and fruit flies. The same group reports that the tetrapeptide reproduces these effects, including restoring the circadian rhythms of melatonin and cortisol in old rhesus monkeys.

A 2025 study from another group found dose-dependent telomere lengthening in normal human epithelial cells and fibroblasts through up-regulation of hTERT and telomerase, and in breast cancer cell lines through alternative lengthening of telomeres. Earlier conformational work proposed binding sites for the tetrapeptide in promoter regions, including that of telomerase.

Studies from the originating programme report decondensation of chromatin in lymphocytes from people aged 75 to 88, protection of oral mesenchymal stem cells from senescence over long-term expansion, and effects on the differentiation of pluripotent and neural cells.

Most of this record comes from the research group that developed the peptide and its collaborators, and many of the in vivo and clinical claims rest on the pineal extract rather than the tetrapeptide. A 2025 review notes that, despite the volume of biological work, physico-chemical and structural studies remain limited and it is uncertain whether the reported mechanisms are the whole picture. The compound holds no FDA approval.
