Three characterised lines of work

Ipamorelin operates by stimulating the pituitary gland to produce growth-related secretions in animal studies. Additionally it has been observed to reduce the production of somatostatin a secretion that can inhibit growth hormone release. Research also indicates that Ipamorelin may enhance the production of IGF-1 (Insulin-like Growth Factor 1) which is associated with processes involved in the growth and repair of muscular and skeletal tissues.

Secretagogue receptor route

The original characterisation describes a pentapeptide identified within a series of compounds lacking the central Ala-Trp dipeptide of GHRP-1. In primary rat pituitary cells it released growth hormone with potency and efficacy reported as similar to GHRP-6, at an EC50 of 1.3 nmol/l against 2.2 nmol/l. Profiling with GHRP and GHRH antagonists indicated that release proceeds through a GHRP-like receptor rather than the GHRH receptor, which is the basis on which the compound is separated from the GHRH analogues sold alongside it.

Potency across species

The same report describes dose-response work in two animal models: in pentobarbital-anaesthetised rats an ED50 of 80 nmol/kg with a maximum of 1545 ng growth hormone per ml, and in conscious swine an ED50 of 2.3 nmol/kg with a maximum of 65 ng/ml plasma. Both are given against GHRP-6 measured in parallel, and both are reported as comparable to it.

Later receptor-level work

A 2024 study examined two growth hormone secretagogue receptor 1a agonists in ferrets given cisplatin, reporting that both anamorelin and ipamorelin inhibited the weight loss and reduced feeding seen in acute and delayed phases. The anti-emetic effect described in that work is attributed to anamorelin alone and to a central mechanism, with the two compounds also compared on electrically stimulated contractions of isolated ileum.

The characterisation this record rests on is preclinical and dates to 1998, in rat pituitary cells and in two animal species. A 2020 review of growth hormone secretagogues in the management of body composition states that clinical efficacy data for this class largely remain lacking, and a 2026 review of peptides used in sports medicine catalogues the compound among those marketed to patients without regulatory approval. The compound holds no FDA approval.
