Three characterised lines of work

Synthetic analogue of growth hormone-releasing hormone (GHRH) developed as a truncated 29-amino acid sequence. This modified peptide exhibits altered receptor binding characteristics and pharmacokinetic properties compared to native GHRH, making it a valuable tool for laboratory investigations.

Two forms under one name

The published human pharmacology of CJC-1295 concerns the form carrying a drug affinity complex: a maleimido group at the C-terminus that binds covalently to plasma proteins such as serum albumin, producing conjugates with a much longer half-life than the unconjugated peptide. Anti-doping work treats the two as separate analytes and identifies their metabolites separately. The form sold here is the one without that complex, and the multi-day duration reported below belongs to the other.

Human dose-ranging trials

Two randomised, placebo-controlled, double-blind ascending dose trials of 28 and 49 days were run in healthy subjects aged 21 to 61, given the compound subcutaneously as single ascending doses in the first study and as weekly or biweekly doses in the second. After a single injection the report describes dose-dependent increases in mean plasma growth hormone of two- to ten-fold sustained for six days or more, with parallel increases in insulin-like growth factor I.

Serum protein profiling

A separate study set out to find biomarkers of growth hormone action, on the stated grounds that the existing markers show substantial variability in the population and do not reliably predict either the effects of therapy or detection of abuse. Sera from 11 healthy young adult men before and one week after injection were compared by two-dimensional gel electrophoresis, with the proteins that changed then identified by mass spectrometry.

The distinction in the first panel is the main limitation of this record: the trials describing several days of raised growth hormone were run on the drug affinity complex form, which binds albumin, and not on the form supplied here. Administration of GHRH analogues is prohibited by the World Anti-Doping Agency, and a 2021 detection study notes that despite evidence of use from admissions and intelligence, these compounds do not appear to have been found in accredited laboratory samples. The compound holds no FDA approval.
